You can big block just about anything, however the newer of the Big Three will have the bolt on kits from the aftermarket sources, and these can really be a time saver. Dooms is correct, it's tough and expensive to make big HP out of small motors but there are some on here who have done it. And the Hi-Po stuff for the Briggs and Honda V-Twins is amazing, anything you could do for a Chevy or Ford you can now do... I always loved building/rebuilding the big chevy v-8 motors, I think it'd be great to do the same thing to one of these small v-twins.
